Wnt/β-Catenin Signaling Pathway | Overview, Purpose and APC Mutations

Lesson on the Wnt/β-Catenin signaling pathway (also known as simply the Wnt signaling pathway), including an introduction to the pathway, Wnt growth stimulatory factors, β-Catenin regulation and role of the pathway in growth, proliferation and development. Wnt proteins act as The Wnt signaling pathway is critically important in embryonic development, particularly in the development of the heart. As such the Wnt/β-Catenin signaling pathway is highly conserved. However, if mutations occur in proteins in the pathway, such as mutations in the adenomatous polyposis coli (APC) gene, this can lead to inappropriate activation of the Wnt/β-Catenin signaling pathway. NOTE: In the video, I misspoke and said that Beta catenin enters the "mitochondria" - I meant to say it enters the nucleus. Sorry about the confusion everyone. Hope you find the video helpful :) Hey everyone, here is another biomedical lesson, and this one is on the canonical Wnt signaling pathway. In this lesson, you will be introduced to the Wnt signaling pathway, and the purpose of the Wnt pathway. You will also learn the signaling pathway in a step-by-step fashion, starting with how the Wnt/β-Catenin signaling pathway is regulated during inactivation, how cellular beta catenin levels are regulated by the destruction complex, and how the pathway can become activated by the extracellular signaling Wnt proteins, which are growth stimulatory factors. Finally, you will learn what happens when the APC protein in the destruction complex is mutated, leading to inappropriate activation of the Wnt signaling pathway. Notch Signaling Pathway | Purpose and Mechanism

Lesson on the Notch Signaling Pathway. The Notch signaling pathway is a highly conserved and critically important pathway in homeostasis and the development of sensory hair cells and branched arterial networks. The Notch signaling pathway derives its name from the NOTCH receptors, which are activated through ligands located on the cell membrane of other cells (sending cells). The NOTCH receptor is composed of three domains, the extracellular domain, a single transmembrane domain, and a notch intracellular domain (NICD). Upon ligand binding to the Notch receptor, the Notch receptor undergoes cleavage reactions, releasing the NICD into the cytosol. The NICD binds to the complex of proteins, which translocate into the nucleus and activate Notch target genes involved in cellular proliferation, differentiation and apoptosis. Hey everyone! Lesson on the Ran GTPase Cycle and the mechanism of Nuclear Transport involving importins and exportins. The Ran GTPase cycle is critically important for nuclear transportation of transcription factors and other proteins into and out of the nucleus. The RanGTPase cycle induces a Ran gradient, which leads to increased levels of RanGDP in the cytosol and RanGTP in the nucleus. Importins and their cargo, along with RanGDP enter the nucleus through nuclear pore complexes. Once inside the nucleus, RanGDP gets processed to RanGTP. Hey everyone. Heme Catabolism and Degradation Pathway - Biochemistry Lesson

Video lesson on catabolism and degradation of heme from hemoglobin during red blood cell (erythrocyte) hemolysis in a powerpoint, university-styled lecture! Hey guys! In this video I (attempt to) simplify and explain the heme catabolism pathway in a (hopefully easy) step-by-step lesson! I first start by explaining basics of the physiology and function of a red blood cell (aka erythrocyte) and that hemoglobin is released from red blood cells during degradation or hemolysis. I next explain hemoglobin is taken up by the reticuloendothelial system (white blood cells like macrophages) and converted to heme. Heme is then converted to biliverdin and then bilirubin, and the iron moiety of heme is stored in ferritin and hemosiderin in white blood cells. From here I explain step-by-step bilirubin metabolism in liver hepatocytes, conjugation and incorporation into bile in the gallbladder, conversion to urobilinogen and stercobilin and urobilin in the urine. I also discuss the enzymes required for each step of the heme catabolism pathway. At the end of the video, I also provide quick facts regarding erythrocytes, hemoglobin, and bilirubin and the clinical "standard" lab values of blood serum bilirubin. Lesson on the transport, absorption and metabolism of ketone bodies (ketolysis) as a source of energy during fasting and intense exercise. Prolonged fasting, intense exercise and certain dietary modifications (ex. ketogenic diet) lead to increases in blood levels of ketone bodies (beta hydroxybutyrate and acetoacetate), which can be used for energy production by peripheral tissues, including the brain and skeletal muscle. Ketone bodies become an important energy source for the brain during fasting and starvation as the brain cannot use fatty acids as an energy source because the blood brain barrier is impermeable to fatty acids. By utilizing ketone bodies (ketolysis), the body can reduce its reliance on glucose as an energy substrate, maintaining blood glucose levels and allowing glucose to be used by other glucose-dependent cells and tissues, such as erythrocytes. In this lesson, you will learn about the blood levels of ketone bodies during fed states, and throughout times of fasting, intense exercise and when utilizing the ketogenic diet. You will also learn about how ketone bodies are transported out of the liver after ketogenesis, how ketone bodies are transported through the bloodstream, and then finally, you will also learn about how ketone bodies are transported into peripheral tissues, such as the brain and skeletal muscle. Additionally, this lesson details the ketone body catabolism pathway in a step-by-step fashion, describing each enzyme in the pathway, the rate limiting step of the pathway, important co-factors for each of the enzymes and what is produced from the body. And finally, you will also learn how much ATP is generated from both beta hydroxybutyrate and acetoacetate. Galactose Absorption and Metabolism | Leloir Pathway and Galactosemia

Lesson on galactose absorption, metabolism via the Leloir pathway, and the medical condition known as galactosemia. Galactose is monosaccharide, which is typically ingested in the form of lactose (galactose + glucose) or free galactose in certain fruits and vegetables. Upon ingestion and digestion, the galactose is transported into an intestinal endothelial cell through SGLT1 and transported into the blood via GLUT2. Upon entry into a receiving cell, galactose is metabolized by the Leloir pathway to form glucose-6-phosphate, which can be rerouted and utilized in the glycolysis pathway to form ATP and pyruvate. If there are any congenital issues causing deficiency in one of three enzymes in the Leloir pathway, this can lead to dysfunctional galactose metabolism and subsequent high blood levels of galactose, leading to issues with red blood cell functioning and increasing risk of cataract formation. In this lesson, we first discuss the dietary sources of galactose, how galactose is digested and how galactose is transported from the intestinal lumen into an intestinal endothelial cell and out into the bloodstream. Lesson on the Non-Canonical NF-kB Signaling pathway. The Noncanonical NFkB signaling pathway is primarily involved in B cell maturation, lymphoid organogenesis and immune system functioning. This signaling pathway differs from the canonical pathway in terms of cell receptors and cytosolic adaptor proteins. In a constitutional, inactivated state, cell levels of NIK are kept low due to proteasomal degradation. However, when this pathway becomes activated, NIK levels rise, leading to activation of IKKalpha and subsequent formation of an active protein dimer. This active protein dimer translocates into the nucleus and leads to the induction of non-canonical NF-kB signaling genes, which increase B cell survival. Lesson on the glucose-alanine cycle (Cahill cycle), how nitrogenous waste is produced in the muscle, how the skeletal muscle “stores” nitrogen in the form of alanine, and how alanine is transported and processed in a liver hepatocyte. Skeletal muscle metabolism of branched chain amino acids, and degradation of purines and pyrimidines leads to the production of ammonium and other nitrogenous wastes. Since the urea cycle does not occur in muscle, the skeletal muscle cannot process the ammonium directly, and thus the skeletal muscle incorporates the ammonium into alanine and glutamine, which are transported to the liver and processed. ********NOTE: 2 Mistakes have been identified in this lesson. 1) Alanine aminotransferase is abbreviated ALT, not AST... AST is actually aspartate aminotransferase 2) The last step in gluconeogenesis in the liver is performed by glucose-6-phosphatase, NOT glucose-6-phosphate dehydrogenase. Lesson on the Cori cycle, introducing the importance and purpose of the Cori cycle and discussing how the Cori cycle can recycle lactic acid, produced from anaerobic metabolism in skeletal muscle, in the liver to form glucose through gluconeogenesis. Intensive exercise and other hypoxic conditions induce anaerobic metabolism in muscle cells, which leads to the production of lactic acid from pyruvate via the enzyme lactate dehydrogenase (LDH). If this lactic acid is left in the muscle cell, the pH of the muscle cell can decrease and cause detrimental effects to the muscle. Therefore, the lactic acid is transported out of the muscle cell through specific transporters located in the muscle cell membrane. The hydrogen ions from the lactic acid are then buffered by the blood and/or removed from the blood in the form of carbon dioxide. The remaining lactate is transported into the liver, where it can be used as a precursor for gluconeogenesis. Hey everyone! Here is a lesson on the Cori cycle and lactic acid metabolism. In this lesson, you will learn the differences between aerobic and anaerobic metabolism in skeletal muscle, and how lactic acid is produced. You will also learn how that lactic acid is transported out of the cell and into the blood, and how the blood buffers and removes excessive hydrogen ions (protons) produced from lactic acid hydrolysis. The chaperone complex in CMA consists of a variety of proteins including Hsc70 and Hsp90, which have important functions for CMA. CMA function requires not only proper chaperone functioning, but also lysosomal functioning. Decreased lysosomal functioning due to decreased lysosomal acidification, results in suppression of CMA. When the CMA chaperone complex delivers the protein cargo to the lysosome, the protein cargo is bound, unfolded and then translocated through a lysosomal membrane receptor known as LAMP2A. The protein cargo is then degraded within the lysosome via proteases known as cathepsins. LAMP2A is indispensable for CMA functioning and a deficiency of LAMP2A can lead to a condition known as Danon disease. Hey everyone.
