Ion exchange chromatogram This lecture explains about the instrumentation, principle of Ion exchange Chromatography. Ion chromatography is a chromatography process that separates ions and polar molecules based on their affinity to the ion exchanger. It works on almost any kind of charged molecule—including large proteins, small nucleotides, and amino acids. However, ion chromatography must be done in conditions that are one unit away from the isoelectric point of a protein.The two types of ion chromatography are anion-exchange and cation-exchange. Cation-exchange chromatography is used when the molecule of interest is positively charged. The molecule is positively charged because the pH for chromatography is less than the pI. In this type of chromatography, the stationary phase is negatively charged and positively charged molecules are loaded to be attracted to it. Anion-exchange chromatography is when the stationary phase is positively charged and negatively charged molecules (meaning that pH for chromatography is greater than the pI) are loaded to be attracted to it.It is often used in protein purification, water analysis, and quality control. The water-soluble and charged molecules such as proteins, amino acids, and peptides bind to moieties which are oppositely charged by forming ionic bonds to the insoluble stationary phase.The equilibrated stationary phase consists of an ionizable functional group where the targeted molecules of a mixture to be separated and quantified can bind while passing through the column—a cationic stationary phase is used to separate anions and an anionic stationary phase is used to separate cations. Cation exchange chromatography is used when the desired molecules to separate are cations and anion exchange chromatography is used to separate anions. Preventing ion exchanger breakthrough is one critical element of a comprehensive program to protect your equipment against the costs associated with turbine corrosion and boiler corrosion. One of the major challenges associated with preventing ion exchanger breakthrough is estimating deionization resin capacity, and predicting when your deionization resin will exhaust and need to be regenerated. This video shows the weaknesses of traditional ways of estimating remaining deionization resin capacity, and presents a more accurate and reliable way of estimating when regeneration will be needed. Preventing ion exchanger breakthrough is one way to protect against turbine and boiler corrosion. Using traditional methods of measuring sodium, silica and conductivity to monitor water purity are all viable options, but they all have a common disadvantage. They detect resin exhaustion only after breakthrough. This means that contaminants have already reached your process water as it is being detected by your analyzer or sensor, which is too late. The two typical methods for estimating breakthrough before it occurs are based on elapsed time (i.e. how much time has passed since the last regeneration) or totalized flow (i.e. how much water has passed through the ion exchanger). Both of these methods require the assumption that flow volumes and/or amount of deionization resin required are constant. This is not typically the case, resulting in one of two outcomes: Early resin regeneration, which wastes both time and money by not fully utilizing the deionization resin capacity. Unexpected resin exhaustion, which results in contamination of your water cycle chemistry, resulting in potential equipment damage. Two solutions are prepared where a few drops of 0.2 M calcium nitrate is added to distilled water. In the second solution, a zeolite is added that will exchange Na+ ions for Ca2+ ions reducing the water hardness. Upon addition of carbonate ions, the precipitate formed in the solution with zeolites is noticeably less and eventually clears up completely. Hard water occurs when magnesium or calcium is dissolved in water and can form deposits with carbonate or with soap. Reducing the amount of calcium or magnesium ions is the goal for water softening.
